Introduction:Theimpactofarthroscopictemperatureonjointtissuesispoorlyunderstoodanditisnotknownhowmesenchymalstemcells(MSCs)respondtotheeffectsofheatgeneratedbythedeviceduringtheprocessofarthroscopyassistedexperimentalcell-basedtherapy.Inthepresentstudy,weisolatedandphenotypicallycharacterizedhumanbonemarrowmesenchymalstemcells(hBMMSCs)fromosteoarthritis(OA)patients,andevaluatedtheeffectofarthroscopicheatoncellsinsuspensionandpelletcultures.
Methods:PrimaryculturesofhBMMSCswereisolatedfrombonemarrowaspiratesofOApatientsandculturedusingDMEMsupplementedwith10%FBSandcharacterizedfortheirstemness.hBMMSCs(16×10cells)culturedassinglecellsuspensionsorcellpelletswereexposedtoanilluminatedarthroscopefor10,20,or30min.Thiswasfollowedbyanalysisofcellularproliferationandheatshockrelatedgeneexpression.
Results:hBMMSCswereviableandexhibitedpopulationdoubling,shortspindlemorphology,MSCrelatedCDsurfacemarkersexpressionandtri-lineagedifferentiationintoadipocytes,chondrocytesandosteoblasts.Chondrogenicandosteogenicdifferentiationincreasedcollagenproductionandalkalinephosphataseactivity.ExposureofhBMMSCstoanilluminatedarthroscopefor10,20,or30minfor72hdecreasedmetabolicactivityofthecellsinsuspensions(63.27%at30min)andincreasedmetabolicactivityincellpellets(62.86%at10minand68.57%at20min).hBMMSCsexposedto37,45,and55◦Cfor120sdemonstratedsignificantupregulationofBAX,P53,CyclinA2,CyclinE1,TNF-a,andHSP70incellsuspensionscomparedtocellpellets.
Conclusions:hBMMSCcellpelletsarebetterprotectedfromtemperaturealterationscomparedtocellsuspensions.TransplantationofhBMMSCsaspelletsratherthanascellsuspensionstothecartilagedefectsitewouldthereforesupporttheirviabilityandmayaidenhancedcartilageregeneration. |
